Rui Chen Lab Research

Genetics of Disease

Genetics plays important role in human visual disorders that affect more than 30 million people in the United States alone. The Chen Lab aims at characterizing the genetic factors underlying the neural degenerative diseases in the human visual system, including both the inherited to age related retinal degenerative diseases. A combination of next generation sequencing, single cell omics, patient derived iPSC and retinal organoid, mice and non human primate models, and machine learning and deep learning technologies are used.

Inherited retinal degenerations (IRDs) are a collection of severe retina diseases following Mendelian inheritance model. Collectively, about 1 in 2000 people is affected by IRD. Over the last decade, our group has recruited over 7,000 patients with IRDs across the world. A combination of next generation sequencing (NGS) based panel, whole exome, and whole genome sequencing has been conducted. These efforts have led to accurate molecular diagnosis of about 85% of the patients, resulting in the discovered of 15 new IRD associated genes. Continued investigation of underlying genetics of the remaining 1,000 unsolved IRD patients provides excellent opportunities for novel disease gene and mutation discovery.

We also extended our study from Mendelian to complex diseases, such as Glaucoma and age-related macular degeneration (AMD), which affect a large portion of population over 65-year-old. We are taking a combination of genomic sequencing, single cell omics, and high through put functional assay to identify causal gene and genetic variants underlying the risk of these conditions.

The Chen Lab is leading the effort of the constructing the cell atlas of the visual system as part of the Human Cell Atlas (HCA) project. Building on this foundation, we aim at gaining insights of the development and the disease of the visual system as the single cell resolution.

The rapidly developing single cell omics technology are revolutionizing many diverse fields of biomedical research. These exciting cutting-edge technologies provide unprecedented opportunities for characterize the developmental and disease process at the ultimate single cell resolution. We are further developing both experimental and computational analysis tools with primary focuses on the spatial single cell technologies and integrative analysis.

Retina Cells

The retina is the innermost tissue of the eyes of human and most other vertebrates. In the human retina, the major cell types could be further classified into subtypes based on their morphology, physiology features or functions, and molecular markers. In total, over 60 cell types are identified in our dataset, making the currently most comprehensive single-cell profiling of adult human retina.

Therapeutics

The Chen Lab is applying what has been learned from the genetics to develop novel therapeutics treating human retinal degenerative diseases.

We are pursuing multiple strategies in parallel to treat human retinal degenerative diseases. For IRDs with defined genetic mutations, the most straightforward treatment is to restore the wild-type function of the mutated gene. In addition to gene augment therapy, we are developing novel therapy with genome editing approach, including base editing, and prime editing.

As the central nervous system (CNS) has little capacity for spontaneous regeneration, repair, or healing in human. severe damage in the CNS, due either to injury or neural degenerative disease, leads to permanent incapacity in patients who often require lifelong assistance. It has been observed that some species, such as fish and birds, can regrow neural cells after retinal injury and restore visual function. We are developing novel approach to active the intrinsic regeneration capability that is lost in humans that would lead to controlled regeneration of the retina tissue upon treatment.
