CPRIT Cancer Proteomics and Metabolomics

Overview

The Cancer Prevention and Research Institute of Texas is supporting a Cancer Proteomic and Metabolomic Core Facility that provides consultation, instrumentation, technical support and data analysis for cancer research projects. The facility operates as a fee-for-service core with very competitive pricing subsidized by the CPRIT grant. The core facility accepts requests for cancer related research from Baylor College of Medicine faculty as a priority.

Services

The core facility provides the following technology platforms as services. Detailed protocols for each platform and policies for operation can be found with attached links:

MS Proteomics

Mass spectrometry identification of protein complexes and their interacting protein networks and untargeted label-free proteomic profiling. This work is conducted under the direction of Drs. Anna Malovannaya and Sung Yun Jung primarily with a Thermo Fisher Orbitrap mass spectrometers.

Reverse Phase Protein Array

Antibody-based reverse-phase protein arrays (RPPA) to quantitatively analyze specific protein pathways and their activation states with large sample sets (tissue or cells). Approximately 240 validated antibodies are available for RPPA. This work is conducted by Dr. Shixia Huang using an Aushon 2470 robotic protein arrayer and a DAKO high throughput auto-slide immunostainer.

Metabolomics

Targeted measurement of steady-state metabolites (up to 700 known metabolites) untargeted profiling, lipidomics and metabolic flux. This work is conducted by Drs. Arun Sreekumar and Nagireddy Putluri using multiple Agilent mass spectrometers and a SCIEX TOF mass spectrometer.

Multi-Omics Data Analysis Core

Consultation on design of projects and data analysis are also provided as Core Services. Data analysis is performed with customized software tools developed for each of the three technology platforms.

Project Requirements

All projects will require submission of a two-page request for project (RFP) application that will be reviewed for feasibility, appropriateness for the available technology platforms, cancer relevance and scientific merit. Approved RFPs will be prioritized, scheduled accordingly, and specifics of each project will be designed prior to initiation. The PI of the requesting lab is required to attend the initial planning meeting of the approved project. Pricing for projects will be provided upon request. At the completion of each project an additional consultation will be provided to review the analyzed data.

Publications that result from the work of the CPRIT Core facility require acknowledgement of the CPRIT Core Facility grant (Proteomics and Metabolomics Core Facility RP 170005).
