OPT (Optical Projection Tomography) is similar to microCT but instead of irradiating the sample with X-rays, light is used either in transmission mode (white light) or fluorescence mode. Optical sections are generated by computed tomography from images taken every 0.3° rotating around 360°. The OPT system images tissue cleared in BABB and other clearing agents and has the ability to generate optical sections from large samples such as an E11.5 mouse embryo using fluorescence.
