PML/RARA Qualitative Analysis
Test Information: Detection of the presence of PML/RARA L-Form (bcr1), S-Form(bcr3), and the rare bcr2 transcripts resulting from t(15;17) translocation in patients with APL.

Special Notes: Technical Limitations:

The analytical sensitivity is equivalent to the detection of 1 fusion transcript positive cell in a background of 1,000 fusion transcript negative cells for L-Form transcripts and 1 in 100 for S-Form transcripts. Absence of a detectable PML/RARA rearrangement does not exclude the possibility of the presence of low levels of transcript not detectable by this assay, or a variant outside the analyzed breakpoint regions.
Results of this test must always be interpreted in the context of relevant clinical and pathological data, and should not be used alone for a diagnosis of malignancy. This test is not intended to detect minimal residual disease.

Methodology: Technical Limitations:

Total RNA isolated from the patient's sample is reverse-transcribed using reverse transcription PCR (RT-PCR) amplification. cDNA generated from RT-PCR is used for PCR reactions using primers that flank the translocation breakpoints of PML/RARA fusion gene transcripts associated with the bcr1 (intron 6, L-Form), bcr2 (Exon 6), and bcr3 (Intron 3, S-Form) breakpoints. The PCR products are evaluated using agarose gel electrophoresis. The assay is designed to detect fusion transcripts described above only. The analytical limitation is 1:1,000 for the L-Form and 1:100 for the S-Form.
