About the Core

The DNA Sequencing Core performs Sanger DNA sequencing on a 3130XL Genetic Analyzer of Applied Biosystems. The core has served the Texas Medical Center research community for more than 22 years with outstanding reputations. Our services include:

The DNA Sequencing Core is open 8 a.m. -5 p.m. every working day. Samples are collected and processed by noon everyday. Results are reported daily, including Saturday.

Content

1. Sample preparation

All templates must be purified to ensure quality of sequencing. Using an agarose gel to verify the quality and quantity of your template (after purification) is the best method. Samples need to meet the requirements below to guarantee a good outcome.

a. Primer: 10 pmol/ul. Please submit at least 1ul/reaction. The core provides the common universal primers such as M13-forward, reverse, T7, T3, Sp6, etc.

b. DNA template: please submit at least 5ul/reaction.

2. Submission

a. Place templates and primers in individual 1.5ml tubes. For multiple reactions to be done on the same template, place enough sample in one tube. Do not pre-mix your sample with primer. Samples will be discarded three weeks after results are sent unless the users request to keep them.

b. Drop off sample at ABBR R651 cabinet with service request sheet by 12 p.m. Sample drop off after 12 p.m will be stored and processed the next working day.

3. Results

Data will be sent by email on the next morning, including Saturday.

4. Data Interpretation and troubleshooting

After verified by core technician, if DNA templates match the quality and quantity requirement, the core voluntarily repeat the sequencing reaction with optimized condition without charge.
