Affymetrix SNPs 6.0 Arrays
Single Nucleotide Polymorphisms (SNPs) and Copy Number Variants (CNVs) play major roles in common human diseases, such as Alzheimer’s, autism, and cardiac and immunologic diseases. Affymetrix’s Genome-Wide Human SNP Array 6.0 contains more than 1.8 million markers for genetic variation and includes probes for SNPs and CNV detection. The near-saturated SNP Array 6.0 allows researchers to eliminate the traditional two-stage SNP studies, cutting down on time by months and decreasing the occurrence of false negatives.
Utilizing the Genome-Wide Human SNP 6.0 Assay, total genomic DNA is digested with restriction enzymes Nsp I and Sty I in separate reactions. Ligators are adapted to the fragments and generic primers that recognize the adaptor sequence bind for the initiation of PCR. The PCR process is optimized to amplify fragments that are 200-1100bp in size. PCR products are subsequently combined, purified, fragmented, labeled, and hybridized to SNP arrays.
