BCR-ABL1 Tyrosine Kinase Domain Sequence Analysis
Test Information: Detection of point mutations in the ABL1 kinase domain of BCR-ABL1 major fusion gene transcripts (p210) in patients with CML.

Special Notes: For initial diagnosis or patients with no previous BCR-ABL1 testing performed, BCR-ABL1-Qualitative test (#9070) should be ordered first. Specimen must arrive in the laboratory within 24 hrs of collection.

Technical Limitations:

The assay is designed to detect mutations within codon 232 to 365 of ABL1 kinase domain of BCR-ABL1 p210 fusion gene only.

Results of this test must always be interpreted in the context of relevant clinical and pathological data, and should not be used alone for a diagnosis of malignancy. This test is not intended to detect minimal residual disease.
Technical Information
Methodology: Total RNA isolated from the patient's sample is reverse-transcribed using reverse transcription PCR (RT-PCR) technology. Nested PCR is performed using the cDNA generated from RT-PCR. The first pair of primers is used to detect BCR-ABL1 fusion gene transcripts p210, and the second pair of primers is used to amplify ABL1 kinase domain (codon 232 to 365). Direct sequence analysis of the second PCR products is performed in both forward and reverse directions using automated fluorescence dideoxy sequencing methods.
