Scheduling

All scheduling of services from the GEM Core should be done through MEMS

Mouse Embryo Manipulation Services (MEMS)
Lab Member (LM) Use
MEMS is a web-based system used to request the services that the Baylor Transgenic Core offers. Once you have registered, have an ID and a password, you have the ability to request Transgenic Core services on-line. Once a service request is in the MEMS system a Request Number is generated specific to the request. It is unique. The request number allows you to follow the progress of the request. The Transgenic Core schedules all requests. The Transgenic Core no longer accepts any request that does not come through MEMS.

To log onto MEMS type in the following address after http://
cb-apps01.cellb.bcm.tmc.edu/mems
Type in your ID and password (I have e-mailed these to you). You may change your password.
Once you are in the system you will see 4 buttons: List View, New Request, Help and Logoff. They are described below.

List View- Lists the requests the LM put in the system. To track requests by the request number, click on the request ID# to view the request details. The request number is unique to your request. When we transfer the mice to you, the Request number will be on the cage card. Please keep track of your requests. You will only have access to the requests you submit.

New Request - Click on the service of choice. Embryonic Stem Cell injection, Microinjection DNA, Transgenic Core Cryopreservation and Transgenic Core Re-derivation are currently offered. Choose the service and fill out the form. When you have filled out the form and submitted it, a unique identifying number will show up at the top of your request. Use the request number to track the progress of your request. PLEASE LABEL the DNA, ES Cell, or Oviduct tubes with this number. Fill out a form for EACH request. Do not put multiple requests on the same form even when they are for the same service. A new request must be made when a service is repeated for any reason. The request number is associated with a particular request done on a particular date.

Please note that for Microinjection you have the option of choosing the Transgenic Core to isolate the DNA for you. Cut 100 ug of DNA in 300 ul of buffer and give us the digest with a picture of a sample cut so we know which band to isolate. Bring the digested DNA to Dr. DeMayo's lab, M725 taped to a copy of the numbered request

The Forms
ALL screens come up pre-populated with your lab information. When a PI makes a request, the PI is listed as Recipient Investigator and Lab Contact. LM's are listed as Lab Contacts when they make requests. Your PI accounts are listed. Check with your PI when you pick an account. The account chosen is the one that is billed for the request. A brief description of how to fill out the forms follows.

Embryonic Stem Cell injection
1. Select account number
2. Type in the CCM assigned project number (not the APRC #)
3. Type Animal room where your mice are to be housed.
4. Type in your APRC#
5. Select the number of clones to be injected
6. Fill out the clone information
7. Indicate in the notes when your clones are available for the core to schedule

Click on Continue
Review the screen information
Click on Submit New Request if the information is correct or click on
Cancel to make changes

When you click on Submit, the unique Request number will appear at the top of the screen. Please make note of this number. Please refer to this number when you have questions.

Microinjection DNA

1. Select account number
2. Type in the CCM assigned project number (not the APRC #)
3. Type in Gene Name (if this is BAC DNA indicate so in the Gene Name)
4. Select the name of the person who prepared the DNA. Select Transgenic Core if we are going to prepare the DNA
5. Type in the DNA preparation method. If the Core is going to prepare the DNA type in Gene Clean.
6. Select animal facility where your mice are to be housed.
7. Select animal room where your mice are to be housed.
8. Type in your APRC#
9. Select strain of embryo to inject.
10. Choose Line of Mice or Transients (choose transients only if you want to take the pups early or have reason to believe the pups may die soon after birth)
11. Enter the OD information if your lab is preparing the DNA. Leave this section blank if the Transgenic Core is preparing the DNA or if it is BAC DNA.
Click the Continue button
Review the request
Click on Submit New Request if correct.
Click Cancel if the information is not correct.

When you click on Submit, the unique Request number will appear at the top of the screen. Please make note of this number. Please refer to this number when you have questions. Print a copy of the request and tape the DNA to it. Bring them to Dr. DeMayo's lab, M725.



Cryopreservation
1. Select account
2. Type in the CCM assigned project number (not the APRC #)
3. Type in Gene Name

Click on Continue
Review the Request
Click on Submit New Request if correct.
Click Cancel if the information is not correct.

When you click on Submit, the unique Request number will appear at the top of the screen. Please make note of this number. Please refer to this number when you have questions.


Re-Derivation
1. Select account number
2. Type in the CCM assigned project number (not the APRC #)
3. Type in animal room where mice are to be housed
4. Type in your APRC#
5. Type in the Gene Name

Click on Continue
Review the Request
Click on Submit New Request if correct.
Click Cancel if the information is not correct.

When you click on Submit the unique Request number will appear at the top of the screen.
Please make note of this number. Please refer to this number when you have questions.
