Cell Based Assay Screening Service
The C-BASS core makes available to the BCM community the licensed GIPZ human and mouse shRNA library collections. C-BASS can provide shRNA clones individually or in pools for use in any BCM laboratory, or carry out an entire screening experiment using the libraries.
C-BASS personnel are available to facilitate the use of these reagents and perform RNAi screens with a level of involvement that is appropriate for the specific experiment. Downstream analyses of the RNAi screen can include, in-house, high-throughput image analysis and flow cytometry. In addition, the identification of the barcoded shRNA plasmids can be carried out at the BCM Microarray Core Facility.
Services and Resources
****************************************************************************************************************
Please read the following information carefully and note the changes made to shRNA clone orders.
Beginning Nov. 1, 2011, C-BASS has set up several interfaces to help with ordering and services. This affects how users communicate with C-BASS regarding products and services. Please check below to ensure you have the most up-to-date information.
- For pricing information please see the C-BASS Price Schedule.
- For reservations of equipment use, please go to http://www.cbass.bcm.tmc.edu/Reservations/. This allows you access to IMXMicro, IMX Ultra, Biomek automated workstations (FX, NX, 3K), and LSRII with HTS. Register first if you are a new user. You will receive email notifications once reservations are complete.
-
For shRNA orders of individual GIPZ clonesIf you have never used our LIMS-based ordering site before, pleaae first go to https://lims.cbass.bcm.edu/lablink/ Please "Request a User ID", and set up your account with your email user name and password. The approval of your account should be within the same day (if request is submitted before 3pm), if you are unable to log in by the end of the day, send an inquiry to c_bass@bcm.edu. Then follow these steps:
FIRST go to https://lims.cbass.bcm.edu/spreadsheet/ to generate a spreadsheet of your search results as an exported Excel file
Then go to https://lims.cbass.bcm.edu/lablink/. Submit your sample sheet after logging into the site.
Please contact c_bass@bcm.edu with any questions.
PLEASE BE ADVISED THAT YOU MAY HAVE TROUBLE ACCESSING LABLINK IF YOU ARE OUTSIDE OF BCM NETWORK. LET US KNOW IF THIS IS THE CASE.
See Ordering Page for more instructions.
-
shRNA orders may be delivered as glycerol stocks or in other formats (e.g., virus). Indicate so during your search. Please see price schedule for rates.
- For all other orders and services, please e-mail c_bass@bcm.edu or call 713-798-8987.
***********************************************************************************************************
1. Clone distribution **The shRNA reagents are available to BCM researchers ONLY**.
- The core distributes licensed human and mouse GIPZ shRNA library collections (that represent a subset of all available shRNA libraries from Open Biosystems), for a nominal fee.
- Each clone – in glycerol stocks – costs $35.
- Orders are normally processed each Wednesday. Orders placed before 5pm on Tuesday will be processed for that week.
- Positive control clones (GAPDH and EG5) and negative control clones (non-silencing and empty vector) are also available for $35 each.
- Information about the new ordering procedures will be updated soon.
2. Target-based shRNA libraries or druggable genome collections
- Kinome and Phosphatome sub-libraries are now available.
- C-BASS anticipates great interests in sub-library collections that are tailored specifically to the interests of the individual investigator. C-BASS is equipped to selectively array a small collection of shRNAs based on your preferences. Examples include shRNAs against a particular signaling pathway or activity. These collections should prove especially attractive to investigators who are more interested in genetically probing the function of a definable set of targets.
3. shRNA-expressing cell line(s) and Lentiviral Stocks
- The C-BASS core will establish the cell line of interest (to be provided by the user) that expresses the shRNA(s) of interest (from the shRNA collection). Lentivirus can be made by the C-BASS core for any of the shRNA or a construct of your interest. For pricing information please see the C-BASS Price Schedule. Discount for bulk orders is available.
4. DNA purification
Facilitated by Beckman Coulter robotics systems, the core will purify DNA in a high throughput manner. 96 well plates will be purified and the OD taken to ensure quality. Contact us for more information.
5. Flowcytometer
The C-BASS core will allow for usage of its BD Biosciences LSRII flowcytometer utalizing only the HTS (high throughput system) for 96 well applications. For more information concerning usage of the LSRII please contact C-BASS.
6. Automated Microscopy
The C-BASS core will allow for usage of its automated microscopes for high-throughput imaging.
